Pharmacokinetics, dosage regimen and in vitro plasma protein binding of intramuscular levofloxacin in buffalo calves

The pharmacokinetics of levofloxacin following its single intramuscular administration (3 mg/kg) was investigated in six male buffalo calves. Peak plasma level of 2.95 ± 0.13μg/ml was observed at 1 h and the drug level above MIC90  in plasma was detected up to 12 h of administration. The bioavailability was 68.1 ± 5.4% and levofloxacin was bound to the plasma proteins to the extent of 19.1 ± 1....

Understanding and reducing the experimental variability of in vitro plasma protein binding measurements.

The experimental measurement of plasma protein binding is a useful in vitro Absorption Distribution Metabolism and Excretion(ADME) assay currently conducted in both screening and definitive early development candidate modes. The fraction unbound is utilized to calculate important pharmacokinetic (PK) parameters such as unbound clearance and unbound volume of distribution in animals that can be ...

Alpha-1 antitrypsin, retinol binding protein and keratin 10 alterations in patients with psoriasis vulgaris, a proteomic approach

Objective(s):Psoriasis is an autoimmune disease that appears on the skin. Although psoriasis is clinically and histologically well characterized, its pathogenesis is unknown in detail. The aims of this study were to evaluate the proteome of psoriatic patients' sera and to compare them with those of normal healthy human to find valuable biomarkers. Materials and Methods: In a case-control study,...

The Binding of Myoglobin by Plasma Protein

When added to dog plasma in vitro and in vivo, myoglobin was bound to plasma protein in a concentration which, maximally, averaged 21 +/- 6 mg. per cent. Electrophoretically, bound myoglobin was separated from free myoglobin and migrated between alpha-2 and beta globulin. The electrophoretic characteristics of protein-bound myoglobin were similar to, although not identical with, those of protei...
